Colin, 2nd Lord Campbell married Elizabeth Stewart and acquired the "Lands of Glume." Castle Campbell was the main Lowland stronghold of the Campbell Earls of Argyll from the fifteenth century until 1654. Inverary Castle is the principal seat of the Campbells .

Colin Campbell, first Earl of Argyll, was Master of the King's Household,Justiciar of Scotland and in 1483 Lord High Chancellor.

In 1489 the Earl persuaded James IV to pass an Act of parliament to change the castle's name to Castle Campbell.

John Knox preached in the south-west corner of the castle, now called "John Knox's Pulpit." The Marquis ( 8th Earl ) of Argyll supported Cromwell and as a stronghold of the Covenanters, Castle Campbell was attacked by the forces of James Graham, Marquis of Montrose who supported the Catholic King Charles I . However the castle withstood the assault. In September 1650, Oliver Cromwell defeated the Scots at the Battle of Dunbar. The castle was taken by Cromwell's forces and used as a garrison . They deserted it and set fire to it in 1654. The Marquis ( 8th Earl ) of Argyll was executed, and the Campbells abandoned their castle in favour of Stirling Castle.

The tower has very thick walls with few windows and one of its original two entrances. The south range was added to Castle Campbell and, about one hundred years later the castle was extended again with an east range.
Archibald, fourth Earl of Argyll, distinguished himself fighting the English at the Battle of Pinkie in 1547 and at the siege of Haddington a year later. In 1563 Mary Queen of Scots stayed at the castle to attend the wedding of the fifth Earl's sister to James Stewart,Lord Doune.
